Abstract:

There are many evidences which demonstrated that the acute ingestion of high concentration of alcohol ha a dirct injurious effect on gastric mucosa . Multiple mechnaismo have been implicated in the pathogenesia of ethanol-damage include the roicrovasculer-ischeraic injury that may result d¥e to""""the release of the chemical mediators like hidtaraine and l^xrKotrine (LTC4) .Also there are -s-trpng evid ,ncee of the " "po'sslbi 1 ity of"oxygen-der: .ved free-radicals formation in large number due to ethanol These observation raised The question about the effect of the conventional doses of some drugs which may reduce these injurious mechanisms and produced a possible gastric cytoprotective effect in addition to their primary action for which they have been used . In this study gastric mucosa lesion parameters , changes in the oxygen-derived in serum glucose . calcium , phosphate were measured . Also the possibility to have any changes in the serum trace elements zinc and copper levels following alcohol administration have b«en measured all these parameters were investigated following the administration of some drugs and their results are summarized as below : 1) Ethanol (2.5 mis of 50 % concentration was found very effective to produce 100 % induction of acute gastric lesions which occur only in the glandular portion of the stomach . This is associated with a significant rise in the levels of oxygen-derived free radicals in both gstric mucosal tissue and serum . Serum glucose and calcium levels showed a non-significant increase which no significant change occur in the phosphate level Both copper and zinc showed a non-significant reduction in there levels . 2) OmBeprazole (10 mg/kg given orally 1 hour before ethanol) produced a significant reduction in ulcer parameter with a preventive index equal to (96.9 %) also a significant reduction in the free - radical levels in both gastric mucosal tissue and serum suggesting a possible cytopro-tective property of omeprazole this effect was associated with a non-significant increase in serum glucose , calcium and phosphate while serum trace elements remained unchanged. 3) The dihydropyridines ca Icium channel Mockers amlodipine (2.5,5mg/kg) , Nifedipine (5 , 10 mg/kg) , Nimodipine (15 Mg/kg) given orally 1 hour before ethanol , produced a significant reduction in all lesions parameters,These reduction have been found to be dose dependant , the preventive index of each one of them was , (84,95 %) for nimodipine . (79 , 92%) for nifedipine and 69% for nimodipine . Aleo amlodipine and nifedipine produce a significant reduction in free radical levels in both
