Abstract:

In this study, microspore developmental stages of Solanum melongena L. Ishtar F1 hybrid were determined cytologically. The size and appearance of flower buds and anthers were determined morphologically. Cold pretreatment was applied to flower buds for (24, 48 or 96 hrs.) in order to induce the androgenetic ability. High percentage of callus induced on untreated anthers grown on Pelletier To medium (cited from Isouard et al., 1979) supplemented with 4 mg/l NAA and 1 mg/l kin. Different levels of kin were used for the induction of organogenesis in callus cultures obtained from anthers of control and cold pretreated flower buds. Callus tissues initiated roots when 0.1 mg/l kin was supplemented to the Pelletier To medium. Incubation of anthers on Pelletier To medium containing 4 mg/l NAA and 1 mg/l kin in continuous dark gave high percentage (10%) of callus induction. Using C medium (Dumas de Vaulx et al., 1981) containing 5 mg/l of 2, 4-D and kin, high percentages of callus (10%) and normal-looking embryos (3.3%) obtained on anthers incubated in continuous dark after transfer to R medium (Dumas de Vaulx et al., 1981) supplemented with 0.1 mg/l kin. Two sucrose levels (30 and 120) g/l were used. On 120 g/l, maximum induction of callus (5%) and normal-looking embryos (3%) occurred on anthers grown on C medium supplemented with 5 mg/l of 2, 4-D and kin then transferred to 0.1 mg/l kin. Different concentrations of activated charcoal (A.C.) (0.5, 1.0 or 1.5) g/l were supplied to C medium containing 5 mg/l of 2, 4-D and kin and R medium supplemented with 0.1 mg/l kin. High percentages (6.6%) of callus induction were obtained on anthers grown on 1.0 g/l A.C., while (3%) normal-looking embryos on 0.5 g/l A.C. Plantlets regeneration (3%) occurred on 0.5 g/l A.C. Rooting of plantlets carried out on V3 medium (Chambonnet, 1985) supplemented with 2% sucrose, 0.5 g/l A.C. and no PGRs were added.
